DNA BARCODE IKAN BUJUK (Channa lucius) ASAL SUNGAI RAWAS DAN SUNGAI KELEKAR BERDASARKAN GEN COI
Forest Snakehead (Channa lucius) is one of the fish that has economic value. The morphological approach is still not effective in determining fish spesies this is because some fish species have similar body shapes, therefore it is necessary to conduct genetic approach using DNA barcode. The pupose of this study was to determine the morphometrics and meristics of forest snakehead, obtained the Cythocrome C Oxydase Subunit I (COI) mtDNA gene sequence, to know the percentage of nucleotides, genetic distance and phylogenetic of the forest snakehead.The methods used in species barcoding and sequence variation were DNA isolation, electrophoresis, amplification using PCR (Polymerase Chain Reaction) and sequencing of COI mtDNA gene region from fish samples obtained from the Rawas and Kelekar rivers. This research was conducted from October – November 2020. The morphometric and meristic result forest snakehead from the Rawas and Kelekar rivers are relatively same. The length of COI gene of forest snakehead nucleotides was approximately 726 bp. The water qualites in the Rawas and Kelekar River were temperature 26.5 – 32.0ºC, brightness 30.5 – 50.0 cm dissolved oxygen (DO) 4.83 -6.89 mg/L, pH 5.6 - 8, ammonia 0.014-0.098 mg/L, alkalinity 7 - 24 mg/L, and total dissolve solid (TDS) 10 - 32mg/L.
