The Effect of Incubation Time on Quality of Semen Simmental Cattle Sexing BSA (Bovine Serum Albumin) Column Method (Supervised by LANGGENG PRIYANTO). This study aims to determine the effect of incubation time on semen quality Simmental cows sexing result of the BSA (Bovine Serum Albumin) column method. This research was conducted in October 2020 at the Laboratory of Animal Reproduction and Health, BIBD (Regional Artificial Insemination Center) Sembawa Banyuasin, South Sumatra. This study used a completely randomized design method (CRD) with 3 treatments and 4 replications. The treatments consisted of P1 (30 minutes of incubation time, 5% and 10% BSA concentration with 4 repetitions), P2 (45 minutes of incubation time, 5% and 10% BSA concentrations with 4 times repetition), P3 (60 minutes of concentration incubation time) BSA 5% and 10% with 4 Repetitions). The parameters observed were the quality of spermatozoa, namely fresh semen and semen from sexing results. Fresh semen includes macroscopic tests and microscopic tests. Macroscopic tests that were observed included color, smell, volume, consistency and pH. Meanwhile, microscopic tests include mass movement, motility and concentration. Semen sexing results include sperm motility and viability. The results showed that the motility and viability of sexing the semen of Simmental cattle with the effect of the incubation time of 30 minutes, 45 minutes and 60 minutes of the BSA (Bovine Serum Albumin) column method had no significant effect. The conclusion of this study is that the incubation time during sexing can reduce the motility and viability of spermatozoa.
