DNA BARCODE IKAN LAIS (Famili Siluridae) BERDASARKAN GEN SITOKROM C OKSIDASE SUBUNIT I (COI)
Glass catfish in Indonesia known as Lais, found in Sumatra, Kalimantan and Java. Glass catfish is very important to be conserved and cultured for consumption and ornamental fish.The purpose of this study were to determine the gene sequence of the Cytochrome C Oxidase Subunit I Mitochondrial DNA, to know the percentage nucleotides similarity between species,to construct the genetic distance between species, and to know the chemical physical parameters of the waters of glass catfish habitat in Lematang River, Muara Enim and Kelekar River, Ogan Ilir. The method used in barcoding species and determining phylogenetic were DNA isolation, DNA amplification using PCR (Polymerase Chain Reaction) and sequencing of COI gene regions of mtDNA.This research was conducted from September – November 2020. The length of COI gene of glass catfish nucleotides was aproximately 693 bp. Glass catfish from the Kelekar River indicated the highest percentage with a subcluster Phalaceonotus bleekeri (KF805378.1) originating from Thailand 92.29% and Phalacronotus apogon (EF609377.1) originating from Australia 92.21%, while the sample of glass catfish from Lematang River showed the highest with subcluster Ompok hypophthalmus originating from Riau 91.63% (MK473378.1). The water qualites in the Kelekar and Lematang River were temperature 29.1 – 32.0⁰C, pH 7.1 – 7.5, dissolved oxygen 4.5 – 5.5 mg L-1, water transparency 25 – 40 cm, alkalinity 10 – 25 mg L-1, ammonia 0.16 – 0.57 mg L-1, and TDS 23 – 37 mg L-1.
