Background: Staphylococcus aureus (S. aureus) was reported as a pathogen that cause oral and dental problems. Increased incidences of resistance and side effects of synthetic antibacterial drugs have led to the need on alternative natural antibacterial products. Catfish as a commodity of freshwater fish from Sumatera Selatan was reported contain omega-3 and omega-6 which have potential effect as antibacterial agent. Aim: The purpose of this study was to determine the antibacterial activity of catfish oil against S. aureus. Method: This study was an in vitro laboratory research. The catfish oil was extracted using Bligh and Dyer method, then continued with low temperature crystallization. Antibacterial activity of catfish oil against S. aureus was evaluated using microdilution method to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC), as well as using the disc-diffusion method to determine the zone of inhibition. This study used catfish oil with concentrations of 0.19-100 μl/ml and chlorhexidine 0.2% as positive control. Results: The result showed that all concentrations of catfish oil extract couldn’t inhibit the growth and kill S. aureus based on the cloudy appearance on microplate wells and the presence of S. aureus on agar plate, while for chlorhexidine 0.2% showed a clear appearance and no bacterial growth. Similarly to the disc-diffusion result, all concentrations of catfish oil extract had no inhibition zone, while the inhibitory zone that was formed using chlorhexidine 0.2% was 16.91 mm. Conclusion: Catfish oil extract with the concentrations of 0.19-100 μl/ml had no antibacterial activity against S. aureus.
