PATOGENESITAS ISOLAT JAMUR ENTOMOPATOGEN ASAL SUMATERA SELATAN YANG DIBIAKKAN DI MEDIA CAIR TERHADAP LARVA SPODOPTERA FRUGIPERDA
Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) was a pest originating from America that had spread to other countries and caused many maize yield losses. To control S. frugiperda, entomopathogenic fungi could be used. Entomopathogenic fungi was biological control that could infected insects. Therefore, this study aimed to determined the pathogenicity of entomopathogenic fungi isolates from South Sumatra from maize root soil and infected S. frugiperda larvae from South Sumatra used liquid media culture against S. frugiperda larvae. This study was conducted at Entomotology Laboratory of Plant Pests and Diseases Departement, Plant Protection Study Program, Agriculture Faculty of Sriwijaya University from March to November 2020. This study used isolates obtained from root soil and infected larvae from South Sumatra. Then, the pathogenicity test used a completely randomized design (CRD) used an incubator with 25 ℃ temperature and 97% relative humidity (RH). This study was conducted using 12 treatments which were repeated 3 times and observed for 12 days. The results obtained 11 isolates of Beauveria sp. namely LtTpOI, TaTsOI, TaAlPA, TaSkPA, TaBrPGA, TaCjPGA, LtApPGA, LtKrLH, TaTtLH, TaLmME, TaPsBA bred in GYB (Glucose Yeast Broth) from root soil and infected larvae in South Sumatra. Macroscopically, Beauveria sp. colony had white upper surface and yellow bottom surface. The older colony isolate would be thicker and yellowish white. The highest mortality of S. frugiperda larvae with the shortest LT50 and LT95 were TaAlPA isolates (62.67%) with LT50 and LT95, namely 8.32 days and 16.94 days. The isolate that had the most influence on leaf area eaten was LtApPGA with 5.22 cm2/head on the last observation days. The lowest percentage of imago appearing were TaLmME (25.33%) and LtKrLH (33.33%). The lowest percentage of pupae that appeared came from TaLmME isolates (25.33%) and LtKrLH (33.33%). The highest percentage of abnormal imago came from TaCjPGA isolates (22.22%), and the highest percentage of abnormal pupae came from TaSkPA isolates (26.67%). The lowest pupal weight was TaTtLH (118.33 mg / head) and the smallest pupal length was TaBrPGA (1.20 cm / head). The conclusion of this study was the entomopathogenic fungi that were cultured in liquid media came from the Beauveria genus. The most pathogenic isolate was TaAlPA with the mortality of S. frugiperda larvae (62.67%) and the shortest LT50 and LT95 were 8.32 days and 16.94 days. The second most pathogenic isolate that had S. frugiperda larvae mortality was TaTsOi (62.67%) and the shortest LT50 and LT95 were 8.67 days and 17.30 days.
