ISOLASI SENYAWA TURUNAN TRITERPENOID DARI FRAKSI ETIL ASETAT DAUN TUMBUHAN Ludwigia peruviana DAN UJI PENGHAMBATAN ENZIM α-GLUKOSIDASE
The isolation of triterpenoid from ethyl acetate fraction of Ludiwigia peruviana leaves and α-glucosidase enzyme inhibition test from extract, fraction and the isolated compound had been conducted. The isolation process was conducted by maceration using methanol as the solvent, then continued with liquid-liquid partition using n-hexane and ethyl acetate. Ethyl acetate fraction was separated and purified using vacuum liquid chromatography and gravity column chromatography. The isolated compound was a white solid with 29.8 mg and decomposed at 278 °C. Based on the analysis of UV, IR, 1H-NMR, 13C-NMR, HSQC, dan HMBC spectra and compared to the literature data shows that the isolated compound was a derivatives of triterpenoid compound, namely 3-O-(E)-coumaroylhederagenin. The α-glukosidase enzyme inhibition test showed that methanol extract, n-hexane fraction, ethyl acetate fraction and the isolated compound gave the IC50 values of 45.07, 448.81, 54.68 and 154.45 ppm respectively while acarbose as positive control was 80.77 ppm. When compared to acarbose, methanol extract and ethyl acetate fraction had a stronger ability as an inhibitor of the α-glukosidase enzyme, while n-hexane fraction and the isolated compound had a weaker ability to inhibit the activity of the α-glukosidase enzyme.
