Preparation and Characterization Submicro Particle of Poly (Lactic Co-Gycolic Acid) Loading Salbutamol Sulfate Using Stabilizer Poly (Vinyl Alcohol) Faris Irvan Saputra 08061381621055 ABSTRACT Salbutamol Sulfate belongs to the Class III Biopharmaceutics Classification System (BCS) which shows that its solubility is high but has a low permeability. Low permeability will affect the bioavailability of a drug so the preparation and characterization of Salbutamol Sulfate is a submicro measure to increase its permeability so as to improve its bioavailability in systemic circulation. Preparation was carried out by a single emulsion solvent evaporation method that uses Poly (Lactic co-Glycolic Acid) (PLGA) polymers and Polyvinyl Alcohol (PVA) stabilizers. Variations in PVA concentrations carried out on the three formulas include 1.6% for formula 1, 2% for formula 2, and 2.4% for formula 3. Determination of the percent efficiency of encapsulation (% EE) results for the three formulas include 89.55 ± 0.100% (formula 1), 92.48 ± 0.044% (formula 2), and 90.85 ± 0.088% (formula 3). Formula 2 is characterized by physical properties using a Particle Size Analyzer (PSA) so that the results for formula 2 are particle diameter 295.63 ± 1.50 nm, size distribution (PDI) 0.43 ± 0.08, and zeta potential -1 , 37 ± 0.47 mV Stability Testing Heating Cooling Cycle Test for 12 days obtained the results in the formula 2 are clear and slightly turbid, there is no phase separation, and there is no aroma of ethyl acetate. The solubility test results show submicro salbutamol sulfate-PLGA soluble in SGF and HCl 5%. Keywords: Salbutamol Sulfate, PLGA, PVA, Submicro Particles, Percent Encapsulation Efficiency
