KARAKTERISASI EKSTRAK KOLAGEN DARI KULIT KAKI AYAM (GALLUS DOMESTICUS)
The objective of the research was to analyze the effect of papain enzyme concentration and extraction time on the physicochemical characteristics of chicken feet skin collagen. The research used a Completely Randomized Factorial Design with two factors, the first factor was papain enzyme concentration (factor A) consisted of two treatment levels (3 and 6%), and the second factor was extraction time (factor B) consisted of three treatment levels (24, 48, and 72 hours). Each experiment was repeated three times. The observed parameters were chemical characteristics of chicken feet skin (water, ash, and protein content), physical characteristics of collagen extract {yield, color (L*, a*, b*) and whiteness index}, and chemical characteristics of collagen extract {protein content and fourier transform infrared (FTIR) analysis}. The results of this research showed that the papain enzyme concentration had a significant effect on yield, protein content, and color (b*). The extraction time had a significant effect on yield, protein content, color (L*, b*), and whiteness index. The interaction of those treatments had a significant effect only on protein content. The fourier transform infrared (FTIR) analysis of collagen extract showed absorption peaks in the amide A, amide B, amide I, amide II, and amide III regions identified the structures in the collagen protein. The optimum condition for collagen extraction to produce the best treatment was a combination of 3% papain enzyme concentration and 24 hours extraction time (A1B1). The best treatment characteristics were a yield value of 0.80% (d.b), protein content of 91.44%, had a functional group at the absorption peak in amides A (3273.87 cm-1), amide B (2918.30 cm-1), amide I (1627.27 cm-1), amide II (1541,16 cm-1), amide III (1241.12 cm-1), still had triple-helix structure in amide I and amide III, and whiteness index 58.35%.
