PEMBIAKKAN JAMUR ENTOMOPATOGEN DI MEDIA AGAR DAN PATOGENESITASNYA TERHADAP LARVA SPODOPTERA FRUGIPERDA
Spodoptera frugiperda was pest originated from America which caused significant losses to agricultural crops, especially maize. Maize that attacked by S. frugiperda would suffer heavy damage. One good way to control S. frugiperda was utilized natural enemy, such as entomopathogenic fungi. Many countries had used entomopathogenic fungi to controled S. frugiperda. Therefore, this study aimed to determined the pathogenicity of entomopathogenic fungi cultured in agar against S. frugiperda larvae. This study was conducted at the Entomotology Laboratory, Department of Pests and Plant Diseases, Faculty of Agriculture. Sriwijaya University from March 2020 to November 2020. This study used isolates obtained from exploration in several districts in South Sumatra by taking infected S. frugiperda larvae in the field and soil around maize roots. Furthermore, the pathogenicity test of S. frugiperda larvae used a completely randomized design (RAL) using 12 treatments that were repeated 3 times and put in an incubator with a temperature of 25 ° C and a relative humidity of 97% and observed for 12 days. The results of this study found 11 isolates cultured GYA (Glocose Yeast Agar) with isolate codes LtTpOI, TaTsOI, TaSkPA, TaAlPA, TaBrPGA, TaCjPGA, LtApPGA, LtKrLH, TaTtLH, TaLmME and TaPsBA. Macroscopically Beauveria sp. It had colony characteristics on days 1 to 7, which was grew spread out and had white flour. Microscopically, Beauveria sp. had hyphae that were insulated with round conidia. Isolat Beauveria sp. tested against S. frugiperda is pathogenic with mortality (50.67% -80.00%) and the highest mortality was found in TaPsBA. LT 50 and LT95 7.97 days and 15.35 days were found in TaTtLH. Larvae that became the lowest pupa were in TaPsBA isolate (20.00%) and larvae which became the lowest immago were in TaPsBA (18.67%). The lowest pupa weight was found in TaLmME around 109.33 mg and the lowest pupa length with an average of about 3.79 cm was found in LtApPGA isolates. The lowest wing span in this study was found in TaTsOI isolates for male wing span (1.28 cm / head) and female wing span (1.31 cm / head). The lowest immago body length was found in taCjPGA for both males (1.28 cm / head) and the lowest female imago body was in TaTSOI (1.31cm / head). The conclusion of this study was discover 11 isolates of Beauveria sp. Cultured in agar medium for Beauveria sp. Isolate GYA. The dead S. frugiperda larvae were characterized by stiff death, dried and covered with white mycelia. The most pathogenic isolate that controled S. frugiperda larvae were TaPsBA with mortality reached 80.00% and LT50 and LT95 the shortest were 7.97 days and 15.35 days.
