EKSPRESI PROTEIN REKOMBINAN PEPTIDE YY (PYY) PADA Escherichia coli
Background: PYY (1-36), a polypeptide consisting of 36 amino acid structures, is known to play a role in increasing the growth and survival of beta cells through NPY1R activation in the pancreas, which ultimately leads to glycemic balance. (P3L31P34)PYY(1-36) is a PYY (1-36) analog that is resistant to DPP-IV degradation and has high selectivity to NPY1R. This study aims to identify the characteristics of the (P3L31P34)PYY(1-36) recombinant protein-coding gene and its prediction of homologous proteins. Methods: This study is an experimental study with a computational approach. Data in the form of amino acid sequences were accessed from Pubmed and the iGEM registry. This study uses several website-based applications including OptimumGeneTM, Predict Protein, Protein-SOL, and SWISS-MODEL. Results: Construction 1 was characterized by a secondary structural composition consisting of α-helix and loop, and a solubility value of 0.5. While construction 2, the secondary structure composition contains α-helix, β-strand, and loop with a solubility value of 0.821. In the (P3L31P34)PYY(1-36) modelling, three homologous protein models were obtained. Conclusion: Construction 1 and 2 have different secondary structures. Meanwhile, the solubility value of construction 2 has a higher value. (P3L31P34)PYY(1-36) has three homologous protein models.
