Telah dilakukan penelitian penentuan kondisi optimum aktivitas enzim xilanase
termostabil dari bakteri Brevibacillus thermoruber. Bakteri B. thermoruber
diisolasi dari sumber air panas Tanjung Sakti Lahat. Pengamatan dilakukan
terhadap pertumbuhan B. thermoruber selama 24 jam dan setiap 2 jam diukur OD
(Optical density). Fase pertumbuhan cepat (eksponensial) didapatkan pada jam
ke-2 sampai jam ke-16. Ekstraksi enzim dilakukan dengan cara sentrifugasi dan
diperoleh supernatan sebagai ekstrak kasar enzim. Ekstrak kasar enzim yang
diperoleh diukur aktivitasnya dengan metode DNS dan kadar protein dengan
metode biuret. Aktivitas enzim xilanase tertinggi diperoleh pada jam ke-14
sebesar 3,38 X 10-2 Unit/mL dengan kadar protein sebesar 3,49 mg/mL.
Penentuan kondisi optimum dilakukan melalui variasi waktu inkubasi (3, 4, 5, 6,
7, 8 menit), konsentrasi substrat (0, 0,5, 1, 1,5, 2%), suhu (40, 50, 60, 70, 80oC)
dan pH (7, 8, 9, 10, 11, 12). Berdasarkan hasil penelitian, didapatkan kondisi
optimum pada waktu 6 menit, konsentrasi substrat 1 %, suhu 600C, dan pH 10
dengan aktivitas sebesar 3,32 X 10-2 Unit/mL
