Silver nanoparticles (AgNPs) are used as an alternative way to overcome antibiotic resistance because silver nanoparticles have antibacterial properties. Therefore, this research aimed to synthesize silver nanoparticles with an environmentally friendly method using bark extract of Sendulang (Antidesma ghaesembilla) and antibacterial activity against Staphylococcus aureus and Escherichia coli. The research started by mixing 40 mL of 1 mM AgNO3 with bark extract of A. ghaesembilla in various volumes (3 mL, 5 mL, 7 mL) and heated at various temperatures (50°C, 60°C, 70°C) for 1 hour. The ability of bark extract A. ghaesembilla to reduce Ag+ ions to Ag0 was indicated by the color change in the AgNPs colloid from clear to brownish yellow. The observation was also supported by the UV-Vis spectrum of AgNPs which appeared in the wavelength range of 400-460 nm at 433 nm, which is a typical SPR absorption region of silver nanoparticles. The optimum conditions for AgNPs synthesis were made using variations in extract volume, heating temperature, and incubation time. The optimum conditions obtained were at 5 mL extract volume, 60°C temperature and 240 hours incubation time. AgNPs from the optimum condition was further characterized using FTIR and XRD. The results of FTIR characterization of silver nanoparticles from the optimum conditions show that OH groups acted as reducers and stabilizers of silver nanoparticles. The results of XRD characterization of silver nanoparticles from the optimum conditions showed diffraction angles 2θ of 38.16°, 44.36°, 64.64°, and 77.00° which were in accordance with JCPDS No. 04-0783 data for silver nanoparticles, the crystal size was calculated using the Scherrer equation and obtained a crystal size of 17.67 nm in the form of FCC (Face Centered Cubic) crystals. The results of the AgNPs antibacterial activity obtained a clear zone diameter of 9 mm on S. aureus and 8.5 mm on E. coli with its inhibitory ability in the medium category.
