FORMULASI DAN EVALUASI SEDIAAN ACNE PATCH TRANSDERMAL EKSTRAK ETANOL DAUN CABAI RAWIT (Capsicum frutescens L.) DENGAN VARIASI KONSENTRASI KITOSAN DAN POLIVINIL ALKOHOL
Acne is a condition characterized by the accumulation of sebum from sebaceous glands in the skin layers, which often stimulates the growth of acne-causing bacteria, such as Staphylococcus aureus. Acne treatment can be performed using acne patches; however, commercial acne patches are only capable of absorbing sebum without providing antibacterial activity. This study aimed to enhance therapeutic effectiveness and drug residence time by formulating acne patches using chitosan and polyvinyl alcohol (PVA) as formulation factors in a 2² factorial design, combined with the ethanolic extract of cayenne pepper leaves, which is known to possess antibacterial activity. Four patch formulations were prepared by combining chitosan and PVA at high and low concentration levels. The effects of formulation factors and their interactions were evaluated through tests of weight uniformity, thickness, surface pH, folding endurance, percent elongation, swelling index, and mucoadhesive time. Data were analyzed using Analysis of Variance (ANOVA) to determine the effect of chitosan and PVA, as well as the antibacterial activity of the patches. The results showed that chitosan had a positive correlation with percent elongation and mucoadhesive time, and a negative correlation with the swelling index, but exhibited no significant effect on weight uniformity and thickness. PVA significantly increased mucoadhesive time but had no significant effect on weight uniformity, thickness, percent elongation, or swelling index. Antibacterial activity was confirmed by inhibition zones ranging from 14,5 to 21,26 mm, with increased activity attributed to the presence of chitosan.
