Bronze Featherback (Notopterus notopterus, Pallas 1769) is a species of freshwater fish, found in Java, Sumatra, and Kalimantan, Indonesia. The objective of this research is to identify the COI sequence of mitochondrial DNA gene and analyze the similarity percentage in bronze featherback from the Medak River, Musi district Banyuasin and Kelekar River, Ogan Ilir district, analyze genetic distances and phylogenetic trees between species of bronze featherback with the GenBank data center, as well as analyze chemical physics of the waters of bronze featherback habitat. The research was conducted from October-December 2023. The methods used in barcoding species of bronze featherback were DNA isolation, DNA amplification using polymerase chain reaction (PCR), electrophoresis and sequencing of COI gene areas on mtDNA. The fragment of the COI mtDNA gene from was obtained from PCR at an annealing temperature of 52.3°C for 30 seconds with 32 cycles. The nucleotide length of the COI gene from Kelekar River 640 bp and the Medak River 701 bp. Analysis of BLASTn samples of bronze featherback from the Kelekar River have a similarity rate of 97.65-100% with bronze featherback from Malaysia, Thailand, East Java, South Sumatra (Ogan River), Java and Bali. While the bronze featherback samples from the Medak River have similarity rates of 97.53-100% against bronze featherback originating in Malaysia, Myanmar, Thailand, Eastern Java, Southeast Sumatra, Java, and Bali that are on the Genbank data. The phylogenetics of bronze featherback originating in the Kelekar River and the Medak River form in the same, but different subclusters with bronze featherback from Thailand, East Java, South Sumatra (Ogan River), Java and Bali. The results of the measurement of water quality from the Kelekar River and Medak River were a temperature of 29.6-32°C, dissolved oxygen 2.72-10.9 mg L-1, TDS (Total Dissolved Solid ) 0.02-6.44 mg L-1, pH 4-6.2, transparancy 40-90 cm and amonia 0.036-0.2 mg L-1.
