PENGARUH VOLUME CAIRAN RESUSITASI TERHADAP INFLAMASI, NEKROPTOSIS, DAN MORTALITAS PADA MODEL TIKUS SEPSIS Analisis terhadap marker inflamasi (IL-6, TNF-α, dan CRP) dan MLKL (mixed-lineage kinase domain-like)
Introduction: Sepsis remains a leading cause of mortality in intensive care units. Fluid resuscitation is the cornerstone of management; however, excessive volume may exacerbate inflammation, activate necroptosis, and aggravate organ injury. This study aimed to evaluate the effect of different fluid resuscitation volumes on inflammation, necroptosis, and renal injury in a lipopolysaccharide (LPS)-induced sepsis rat model. Method: An experimental post-test only control group design was conducted on 30 male Wistar rats divided into six groups (K1–P4). Following LPS induction at 10 mg/kgBW intraperitoneally, rats received 0.9% NaCl at volumes of 0, 15, 30, and 60 mL/kgBW. Serum levels of IL-6, TNF-α, CRP, and MLKL were measured using ELISA; renal pMLKL expression was evaluated by immunohistochemistry; and kidney tissue injury was assessed by H&E staining with semi-quantitative scale. Data were analyzed using ANOVA, Kaplan–Meier survival analysis, regression, and PLS-SEM structural modeling. Result: The 15 mL/kgBW group demonstrated the lowest levels of IL-6 (2.74 ± 0.54 ng/mL), TNF-α (65.25 ± 9.64 ng/mL), CRP (0.98 ± 0.12 mg/L), MLKL (644.59 ± 58.06 ng/mL), pMLKL (3.40 ± 0.40), and kidney injury score (1.76 ± 0.11) (p
