Maternal separation (MS), is a commonly used model to study the effects of early life experiences. The offspring are separated from the mother during the early postnatal period, resulting in limited exposure to the mother. This separation can affect cognitive development by inducing prolonged stress and altering brain neuroplasticity. This effect was especially seen in the hippocampus, which is important in spatial memory and cognitive function. This study aims to analyze the effect of MS duration on the spatial memory of male white Swiss Webster mice (Mus musculus L.). The research used a true experimental design with a posttest-only control group design. A total of 28 mice were divided into four groups: control (without MS), and MS groups with a duration of 2 hours, 4 hours, and 6 hours per day for 21 days. Spatial memory function was tested using the Y-maze and T-maze. Data analysis was carried out using one-way ANOVA test and post-hoc LSD test. The results showed that MS had a significant impact on spatial memory. In the Y-maze, the percentage of spontaneous turnover decreased with increasing MS duration, with the 6-hour MS group showing a significant decrease compared to controls (p < 0.05). Similar results were found on the T-maze, with the 4-hour and 6-hour MS groups showing a significant decrease in accuracy in path selection. MS affects the cognitive function of mice, with longer duration causing more significant spatial memory impairment. This research emphasizes the important role of the environment that supports cognitive development in early life.
