Infections caused by antibiotic-resistant bacteria, particularly carbapenem-resistant Pseudomonas aeruginosa (CRPA), have become a significant global health issue. Pseudomonas aeruginosa is a common nosocomial pathogen that often demonstrates resistance to many antibiotics. Given the rising cases of antibiotic resistance, alternative treatments are needed, one of which involves utilizing the potential of plants. Plectranthus amboinicus (commonly known as torbangun or bangun-bangun leaves) has been recognized in the community as a medicinal plant with various therapeutic properties, including antibacterial activity. This study aims to evaluate the antibacterial activity of Plectranthus amboinicus leaf extract against CRPA by measuring the minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and bacterial morphological changes after exposure to the leaf extract. Additionally, the study also aims to identify the classes of bioactive compounds present in the leaf extract. This laboratory experimental study used a dilution method to test the antibacterial activity of Plectranthus amboinicus leaf extract. The leaf extract was prepared using 96% ethanol as a solvent and tested at several concentrations ranging from 15.625 µg/ml to 2000 µg/ml. The testing involved measuring MIC and MBC and examining bacterial morphological changes under a microscope after treatment. Phytochemical testing was also conducted to identify active compounds in the leaf extract. The results of the study showed that the Plectranthus amboinicus leaf extract exhibited antibacterial activity against CRPA with an MIC of 31.25 µg/ml and an MBC of 62.5 µg/ml. Bacterial morphological observations after treatment revealed shape changes, indicating damage caused by exposure to the extract. Phytochemical tests identified active compounds such as flavonoids, saponins, tannins, and terpenoids, which contribute to the antibacterial activity. In conclusion, Plectranthus amboinicus leaf extract has the potential as an antibacterial agent against carbapenem-resistant Pseudomonas aeruginosa (CRPA). Bioactive compounds in the leaf extract, such as flavonoids, saponins, tannins, and terpenoids, play a role in inhibiting bacterial growth and killing CRPA bacteria. This research opens the possibility for the development of herbal-based alternative therapies to combat CRPA infections resistant to conventional antibiotics. It is hoped that this study will contribute to the development of alternative treatments using Plectranthus amboinicus leaf extract as an antibacterial therapy, particularly against carbapenem-resistant bacteria. The results of this study are expected to serve as a reference for further research in addressing the global issue of antibiotic resistance.
