Backgrounds: Escherichia coli is a bacteria that often causes serious infections, such as digestive tract, urinary tract and bloodstream infections. One of the main obstacles in its treatment is the ability of these bacteria to form biofilms, which increase resistance to antibiotics and protect against external influences. Uncontrolled use of antibiotics further exacerbates the problem of resistance, with millions of deaths worldwide due to antibiotic-resistant bacteria. As an alternative solution to overcome antibiotic resistance, the karamunting plant (Rhodomyrtus tomentosa (Aiton) Hassk) is known to have antibacterial activity against Gram-negative bacteria, including E. coli. The active compounds in caramunting leaves have been proven to inhibit bacterial growth and biofilm formation. This research aims to explore the potential of karamunting leaves as an effective therapy in overcoming antibiotic resistance, as well as improving public health. Methods: Descriptive research to test the activity of karamunting (Rhodomyrtus tomentosa (Aiton) Hassk) leaf extract against the anti-biofilm Escherichia coli bacteria. This research was carried out at the Microbiology Laboratory, Biotechnology Laboratory, and Biochemistry Laboratory of FK UNSRI. The thick karamunting leaf extract was then made into three variations of concentration, namely 125µg/ml, 250µg/ml and 500µg/ml. The positive control used was an antibiotic in the form of meropenem and the negative control used DMSO solution. Results: In this study, the Minimum Inhibitory Concentration (MIC) of karamunting (Rhodomyrtus tomentosa (Aiton) Hassk) leaves for Escherichia coli bacteria was a concentration of 125 µg/ml. Conclussion: Karamunting leaf extract has antibacterial activity against Escherichia coli bacteria. Karamunting leaf extract with a concentration of 125 µg/ml is the Minimum Inhibitory Concentration (MIC) which is effective as an antibacterial against Escherichia coli bacteria.
