This study aims to evaluate the potential of ethanol extract of putat leaves (Barringtonia acutangula) in stimulating the growth of probiotic Bifidobacterium longum and test its ability to inhibit the growth of pathogenic Escherichia coli bacteria. Bifidobacterium longum is one of the lactic acid bacteria that are beneficial for human gut health, while E. coli bacteria are known as opportunistic bacteria that cause gastrointestinal disorders. Ethanol extract of putat leaves was obtained through maceration method and characterized for its phytochemical content, including flavonoids, saponins, and tanins which are known to have antibacterial activity. B. longum growth stimulation test was conducted by culturing the bacteria in MRS media to which the extract was added at various concentrations (2%, 5%, 10%, 15% and media control). Growth was observed by measuring turbidity (OD600). Inhibition test against E. coli was performed using disc diffusion method with post-stimulation B. longum culture supernatant. The results showed that putat leaf extract with a concentration of 15% provided optimal growth stimulation to B. longum, as indicated by a significant increase in optical density (p < 0.05). The best antibacterial activity test results in the B. longum metabolite + putat leaf ethanol extract test group with a concentration at 100% concentration with a strong category. The concentration of 4.7% was determined as the KHM value while the concentration of 9.4% was determined as the KBM value. The conclusion of this study is that ethanol extract of putat leaves has the potential as a natural probiotic stimulant that not only supports the growth of B. longum, but also strengthens its ability to inhibit pathogenic bacteria E. coli. This study supports the utilization of local herbaceous plants as raw materials for synbiotic supplements.
