KOMPOSISI ALEL SPESIFIK PADA POPULASI IKAN NILA (Oreochromis niloticus) DI DANAU RANAU BERDASARKAN SNP MARKER
Nile tilapia (Oreochromis niloticus) is one of the important freshwater fish commodities in Indonesia that has various strains resulting from genetic selection. This study aimed to determine the morphological characteristics and species or strain composition of Nile tilapia that are cultivated or caught in Ranau Lake by utilizing Single Nucleotide Polymorphism (SNP) molecular markers through the KASP assay PCR method. The study was conducted from December 2024 to January 2025 in Ranau Lake and the Biomolecular Laboratory of Universitas Sriwijaya. The samples were consisted of 10 samples of culture Nile tilapia (DRB) and 10 samples of capture Nile tilapia (DRA). The research stages included sampling, DNA extraction, PCR amplification, DNA visualization, and detection of species-specific SNP. The results showed that the Nile tilapia population in Ranau Lake has morphological and genetic diversity, both in culture and capture. SNP marker analysis identified the presence of more than one strain or species of tilapia and revealed strong indications of hybridization, as seen from the mixed genotype pattern and overlapping morphometric and meristic characters between groups. The diagnostic allele frequency of capture Nile tilapia was 0.35 and its alternative allele was 0.65. The diagnostic allele frequency of culture Nile tilapia was 0.65 and the alternative allele was 0.35. Water quality in capture (DRA) indicated the value of pH 5.3-7.2, temperature 31.1-32.9°C, TDS 0.21-0.39 mg L-¹, dissolved oxygen 2.0-5.4 mg L-¹, transparency 45 cm, ammonia 0.052-0.156 mg L-¹ and alkalinity 158-226 mg L-¹. Meanwhile, water quality in the culture (DRB) denoted the value of pH 6.6-6.9, temperature 27.4-27.7 °C, TDS 0.13 mg L-¹, dissolved oxygen 5.3-5.4 mg L-¹, transparency 65 cm, ammonia 0.058-0.137 mg L-¹ and alkalinity 72.88 mg L-¹.
