FORMULASI SEDIAAN SPANSULE EKSTRAK ETANOL DAUN PUTAT (Barringtonia acutangula) UNTUK MENSTIMULASI PROBIOTIK Bifidobacterium longum DAN MENGHAMBAT Eschericia coli
Diarrhea caused by Escherichia coli infections remains a significant public health problem in developing countries, including Indonesia. Overuse of antibiotics can lead to resistance, so alternatives are needed. Putat leaves (Barringtonia acutangula) contain the flavonoid quercetin, which has prebiotic and antibacterial potential. This study aims to investigate the effect of putat leaf extract spansules on the growth of Bifidobacterium longum and the inhibition of E. coli. The extract was obtained through 96% ethanol maceration, then formulated into spansules using wet granulation and coating methods. The evaluation included spansule quality analysis, probiotic growth testing using the total plate count method, and antibacterial activity testing using the disk diffusion method. The best formula was Formula 1, containing 4% Eudragit L100 and 1% chitosan, showing optimal physical parameters and drug release, and following the zero-order and Korsmeyer–Peppas (non-Fickian) release models. Formula 1 spansules were able to stimulate the growth of B. longum, although to a lesser extent than the positive control (pure quercetin), and showed weak antibacterial activity against Escherichia coli (inhibition zone 1.166 mm). Based on the test results, formula 1 has potential as an oral prebiotic formulation that supports probiotic growth and provides an inhibitory effect against pathogenic bacteria.
