ISOLASI AKTINOBAKTERIA DARI MEDIA BUDIDAYA DAN SALURAN CERNA IKAN GABUS (Channa striata) DENGAN SISTEM BIOFLOK
The cultivation of snakehead fish (Channa striata) significant potential to be developed. Intensive cultivation of snakehead fish has challenges due to waste accumulation, may cause a decrease in water quality and elevated ammonia levels. The biofloc system is an alternative that can be implemented by utilizing microorganisms that can control water quality. This research aimed to determine the actinobacteria isolates that have the characteristics of producing cellulolytic, proteolytic and lipolytic enzymes that are capable of oxidizing ammonia. The research method consist of two stages, namely the preparation stage, which including sampling, isolation, selection, purification and isolate propagation, while next stage is the characterization of actinobacteria isolate remediation. This study used Completely Randomized Design (CRD) for remediation characterization with 3 treatments and 3 replications. The treatments consist of without addition of isolates (P0), cultivation media addition of isolate SKA1 (P1) and cultivation media addition of isolate SIB1 (P2). The parameters observed in research including enzymatic selection, ammonium selection, total colonies, density colonies and specific growth rate. The water quality parameters that measured during research were ammonia, dissolved oxygen, total dissolved solid and pH. The results of isolation, selection and characterization showed that isolates SKA1 (P1) and SIB1 (P2) are the best isolate which exhibited lipolytic, cellulolytic, and proteolytic enzymatic activity and were able to reduce ammonia levels on snakehead fish media cultivation by 31.92% and 21.50%.
