EFEKTIVITAS FRAKSI AIR DAUN KARAMUNTING (Rhodomyrtus tomentosa (Ait.) Hassk.) TERHADAP EKSPRESI GLUT 2 PADAPANKREAS TIKUS PUTIH (Rattus novergicus) JANTAN GALUR WISTAR MODEL DIABETES MELLITUS
Background: Type 2 diabetes mellitus (T2DM) is characterized by impaired glucose metabolism due to insulin resistance and decreased pancreatic β-cell function. One of the key factors in glucose regulation is the expression of GLUT 2, a glucose transporter that functions as a glucose sensor. Although drugs such as pioglitazone can improve insulin sensitivity, they may cause long-term side effects. Therefore, plant-based therapeutic alternatives, such as the leaves of Rhodomyrtus tomentosa, are worth exploring. This study aimed to investigate the effect of the aqueous fraction of Rhodomyrtus tomentosa leaves on GLUT 2 expression in the pancreas of male Wistar rats with a diabetes mellitus model. Methods: An experimental study with a post-test only control group design was conducted using male Wistar rats induced with diabetes. The rats were divided into five groups: three treatment groups receiving the aqueous fraction of R. tomentosa leaves at doses of 100, 200, and 400 mg/kgBW, a positive control group (pioglitazone), and a negative control group. GLUT 2 expression was analyzed using immunohistochemistry and assessed by the H-Score method. Results: The aqueous fraction of R. tomentosa leaves increased GLUT 2 expression in a dose-dependent manner. Doses of 200 and 400 mg/kgBW showed significant increases, with the 400 mg/kgBW group approaching the expression levels of the positive control. In contrast, the 100 mg/kgBW dose resulted in lower expression than the negative control, possibly due to subtherapeutic concentrations of active compounds or the use of stored biological materials. Conclusion: The aqueous fraction of R. tomentosa leaves has the potential to enhance GLUT 2 expression in the pancreas of diabetic rat models, particularly at doses of 200 and 400 mg/kgBW. Further studies are required to elucidate the molecular mechanisms and validate its clinical applicability.
