ISOLASI BAKTERI DARI MEDIA BUDIDAYA DAN SALURAN CERNA IKAN GABUS (Channa striata) HASIL KULTUR SISTEM BIOFLOK
The cultivation of striped snakehead using the biofloc system is still constrained by high ammonia levels, resulting in poor maintenance of water quality in the culture system. Isolation of bacteria are important steps to obtaining new bacterial strains with the potential to improve aquaculture water quality. This study aimed to identify bacterial candidates from the culture media and digestive tract of striped snakehead cultivated using the biofloc system, which have the potential to provide benefits for striped snakehead farming, particularly in improving water quality and the digestive health of the fish. The study was conducted from January to March 2025, involving several stages: sampling, isolation, purification, selection, propagation and characterization of bacterial remediation capabilities. This study used a Completely Randomized Design (CRD) consisting of three treatments: control (P0), bacterial isolate 1KA from the culture media of striped snakehead (P1), and bacterial isolate 1IB from the intestine of striped snakehead (P2), with three replications in the bacterial remediation characterization. The observed parameters included bacterial density, total bacterial colonies, specific growth rate of bacteria, lysis zone diameter, selection of ammonium-degrading bacteria, Potential of Hydrogen (pH), Total Dissolved Solids (TDS), dissolved oxygen and ammonia levels. The results of the study showed that isolates 1KA (P1) and 1IB (P2) were obtained from enzymatic selection, exhibiting the largest lysis zones on proteolytic and cellulolytic media. Isolate 1KA was able to reduce ammonium by 25% and decreased ammonia in striped snakehead culture wastewater by 27%. Isolate 1IB reduced ammonium by 8% and decreased ammonia in striped snakehead culture wastewater by 12%
