Thrombosis is caused by the formation of abnormal blood clots in arterial or venous vessels, which obstruct blood flow. Conventional thrombolytic therapies, such as streptokinase and alteplase, carry a high risk of systemic bleeding. As an alternative, thrombolytic therapy using gold nanoparticles synthesized with green synthesis approach utilizes breadfruit peel extract (Artocarpus altilis) as a natural reducing agent and polyethylene glycol (PEG-6000) as a coating agent. The extract was obtained using two extraction techniques: maceration and sonication. The results showed that the sonication extract was the most effective in producing optimal nanoparticles, as supported by characterization processes, phytochemical screening and TLC with total phenolic and flavonoid content values of 700.962 mg GAE/g and 81.455 mg QE/g, respectively. Subsequently, the nanoparticle formulation was optimized using two variables; concentration of ethanol extract of breadfruit peel and PEG-6000 polymer using a 2² factorial design with Design Expert® 13. The optimized concentrations of the extract were 5% and 10%, while those of PEG 6000 as the coating agent were 10% and 20%. Six response parameters were evaluated, including stability at 450 nm and 550 nm, photothermal effect, thrombolytic activity, and hemolysis at 4 and 24 hours. An increase in PEG-6000 concentration and a decrease in extract concentration significantly improved stability at 450 nm and 550 nm, increased temperature, enhanced thrombolytic activity, and reduced hemolytic activity. The optimal formula produced spherical nanoparticles with a TEM morphology, particle size of 132.5 nm, PDI of 0.272, zeta potential of -3.98 mV, and FTIR spectrum indicating the presence and interaction of functional groups from the extract and PEG-6000 in the nanoparticles.
