Many small vendors sell food and beverages along the roadside without paying attention to hygiene in their business practices. One of these vendors is sugarcane juice sellers, a drink that is highly favored by the community. Beverages sold near highways are vulnerable to contamination by E. coli bacteria. Several factors contribute to this, ranging from sanitation issues to the serving process for consumers. The aim of this study is to examine the presence of E. coli bacteria and the application of hygiene practices among sugarcane juice vendors along Pangeran Ratu Street in Palembang City. This research employs a qualitative method. The study involved 12 informants, consisting of 4 key informants (sugarcane juice vendors) and 8 general informants (sugarcane juice buyers). Data collection was carried out through in-depth interviews and observations, then processed using source triangulation and laboratory testing with the Most Probable Number (MPN) method. The results show that most sugarcane juice vendors lack proper waste bins; however, they provide separate handwashing stations and equipment washing areas. The water used comes from PDAM (local water supply company), either sourced directly from their selling location or brought from home. The equipment used—such as strainers, collection glasses, and grinding machines—was mostly rusty. None of the vendors wore personal protective equipment (PPE) while selling. The sugarcane juice was served using food-grade plastic. Laboratory testing found no detectable *E. coli* contamination in the sugarcane juice, with a result of 0/100ml, which meets the established safety standards. All informants, both key and general, stated that they had no knowledge regarding microbiological contamination in beverages. In conclusion, while vendors have implemented good selling practices, there is a need to enhance their knowledge of hygiene and sanitation.
