KARAKTERISASI SENYAWA KIMIA TERISOLASI DARI FRAKSI n-HEKSANA DAUN TUMBUHAN KIPAHIT (Tithonia diversifolia) SERTA UJI SITOTOKSIK TERHADAP Artemia salina
This research was conducted to isolate chemical compounds from the n-hexane fraction of Tithonia diversifolia leaves and to perform cytotoxic tests on Artemia salina from the methanol extract, n-hexane fraction, ethyl acetate fraction, and isolated compounds using the Brine Shrimp Lethality Test (BSLT). The isolation of chemical compounds was carried out by extraction using the maceration method with methanol as a solvent, followed by fractionation using n-hexane and ethyl acetate solvents. The separation and purification of compounds continued using vacuum liquid chromatography (VLC), radial chromatography, and Sephadex LH-20 column chromatography. The isolated compound obtained was a white solid weighing 7.2 mg. Based on 1H-NMR spectroscopy analysis, the isolated compound consists of a mixture made up of methyl ester fatty acids and a sesquiterpene lactone. GC-MS spectroscopy analysis showed that the mixture of compounds was dominated by three fatty acid methyl ester compounds, including methyl palmitate with a peak area of 17.04%; methyl 9-octadecenoate with a peak area of 41.54%; and methyl stearate with a peak area of 4.34%. Cytotoxicity testing of methanol extract, n-hexane fraction, ethyl acetate fraction, and isolated compounds from T. diversifolia leaves gave LC50 values of 277.42; 972.03; 62.76; and 51.83 ppm, respectively. These values indicate that the n-hexane fraction has a weak cytotoxic level compared to the methanol extract and ethyl acetate fraction, while the isolated compound has a strong cytotoxic level.
