Type 2 diabetes mellitus (T2DM) is a major global health concern with an increasing prevalence each year. It is characterized by chronic hyperglycemia caused by insulin resistance and impaired insulin secretion. Genetic predisposition plays a key role, as individuals with a first-degree family history of T2DM are at higher risk of developing the disease. Vitamin D is known to influence immune regulation and glucose metabolism. Low vitamin D levels have been associated with increased proinflammatory cytokines such as interleukin-6 (IL-6), impaired glycemic control (reflected by HbA1c), and increased body weight, as indicated by body mass index (BMI). This was an analytic observational study with a cross-sectional design. A total of 50 subjects aged 18–25 years with first-degree relatives diagnosed with T2DM were recruited through consecutive sampling. Serum vitamin D levels were measured using chemiluminescent microparticle immunoassay (CMIA), IL-6 using enzyme-linked immunosorbent assay (ELISA), and HbA1c using fluorescence immunoassay (FIA). BMI was calculated from anthropometric measurements. Data were analyzed using Spearman correlation. There was a significant negative correlation between vitamin D and IL-6 (r = -0.307; p = 0.03), as well as between vitamin D and HbA1c (r = -0.429; p = 0.002). No significant correlation was found between vitamin D and BMI (r = 0.026; p = 0.856). The findings suggest that low vitamin D levels may contribute to increased inflammation and poor glycemic control. Early screening and nutritional intervention in high-risk groups, especially first-degree relatives, may serve as a preventive strategy for T2DM.
