ISOLASI DAN KARAKTERISASI ENZIM α -GLUKOSIDASE DARI BERAS KETAN PUTIH (Oryza sativa L. var. glutinosa)
This study aims to determine the activity of the α-glucosidase enzyme from white glutinous rice (Oryza sativa L. var. glutinosa) and determine the optimum conditions of the enzyme at incubation time, temperature, pH and substrate concentration. The test of the activity of the α-glucosidase enzyme used the substrate p-Nitrophenyl-α-D-Glucopyranoside (pNPG). The kinetic parameters of the α-glucosidase enzyme were determined to obtain the Vmax and KM values based on the Lineweaver-Burk equation. The process of isolating the α glucosidase enzyme from white glutinous rice was carried out starting from the extraction of white glutinous rice flour using buffer, then fractionated using ammonium sulfate salt. The results of the study obtained a crude extract of the α-glucosidase enzyme from white glutinous rice having a specific enzyme activity of 0.023 U/mg with an enzyme activity of 0.039 U/mL and a protein content of 1.665 mg/mL. The results of ammonium sulfate fractionation had the highest specific activity in fraction III (40-60%) of 0.136 U/mg with enzyme activity of 0.048 U/mL and protein content of 0.353 mg/mL. The optimum conditions of the α-glucosidase enzyme from white glutinous rice showed optimum activity at an incubation time of 25 minutes, pH 5.8, temperature 37°C and substrate concentration of 0.015 M with enzyme activity of 0.123 U/mL. The kinetic parameter test of the α-glucosidase enzyme from white glutinous rice based on the Lineweaver-Burk equation had a Vmax value of 0.19 M/min and a KM value of 0.00987 M.
