PENGARUH NAPHTALANE ACETIC ACID (NAA) DAN BENZYL AMINO PURINE (BAP) TERHADAP PERTUMBUHAN EKSPLAN DAUN ROSELLA (Hibiscus sabdariffa L.) SECARA IN VITRO
This research was aimed to determine the effect of Naphtalene Acetic Acid (NAA) and Benzyl Amino Purine (BAP) on the growth of rosella (Hibiscuss sabdariffa L.) leaf explants using in Vitro culture. This research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture UNSRI, Bukit Lama, District. Ilir Barat I, Palembang City, South Sumatra with location coordinates 2°59'23.4 "S 104°43'53.4 "E. The research was conducted from July to September 2023. The study used two growth regulators namely Naphtalene Acetic Acid (NAA) and Benzyl Amino Purine (BAP) with 8 treatments including A1P1, A1P2, A2P1, A2P2, A3P1, A3P2, A4P1, A4P2. The results showed that the provision of NAA and BAP had an effect on callus induction and root emergence. The combination of NAA concentration of 2 ppm and BAP 1 ppm was the best treatment on for callus induction and callus emergence time, where callus could appear one week after inoculation. The application of 3 ppm NAA and 1.5 ppm BAP resulted in the most root growth, with 2 out of 10 explants.
