HUBUNGAN KEKERABATAN FENETIK TANAMAN KUNGKIL DAN MATOA (Pometia pinnata J. R & G. Forst.) BERDASARKAN MARKA Random Amplified Polymorphic DNA (RAPD)
Indonesia is known as a Mega Biodiversity country with a very high level of biodiversity. Indonesia has 40% endemic or native plants such as Matoa plants. There are two types of Matoa, namely coconut matoa and papeda matoa, each of which shows different characteristics. Genetic characterization of germplasm for conservation and utilization of plant genetic resources using five Random Amplified Polymorphic DNA (RAPD) primers, namely OPA 19, OPB 15, OPD 08, OPD 11 and OPD 13 and using 4 research samples consisting of Kungkil leaves and three Matoa variants (red, yellow, and green). The five primers produced 67 polymorphic bands and 2 monomorphic bands. The results of PCR-RAPD amplification using five primers showed a percentage of polymorphic bands of 96% which indicated high genetic variation. The results of this study used the Unweighted Pair Group Method with Arithmetic Average (UPGMA) method, resulting in two main clusters in which red and yellow matoa have the closest kinship relationship, meanwhile Kungkil shows the most distant kinship with the three matoa variants due to differences in genetic profile or nucleotide base arrangement. This study provides insight into genetic variation and the potential for utilizing genetic resources from Matoa and Kungkil plants in the context of conservation and breeding.
