HUBUNGAN KEKERABATAN FILOGENETIK TANAMAN KUNGKIL DAN MATOA (Pometia pinnata J.R.& G.Forst) BERDASARKAN SEKUEN ITS rDN
Matoa (Pometia pinnata J.R.& G.Forst) is a plant from the Sapindaceae family with high genetic variation due to natural cross-pollination. This study aims to analyze the genetic variation and phylogenetic relationships between Kungkil and several Matoa variants (Red Matoa, Yellow Matoa, and Green Matoa) based on Internal Transcribed Spacer (ITS) rDNA sequences. The methods used include DNA isolation, quantitative and qualitative testing, DNA amplification using PCR, DNA sequencing, and phylogenetic analysis using BioEdit and MEGA 11 software. The results showed genetic variation in the ITS rDNA region with mutations at 35 points, consisting of 12 transversion mutations, 17 transition mutations, and 6 deletion mutations. Phylogenetic tree reconstruction revealed that Red Matoa, Yellow Matoa, and Green Matoa clustered together with Pometia pinnata voucher Chase 2135, while Kungkil was in the same cluster as Pometia pinnata voucher A4659. These findings indicate that Kungkil has genetic differences from other Matoa variants. This study provides valuable genetic information for the conservation and breeding of Matoa plants in South Sumatra. Further studies using additional molecular approaches are recommended to enrich the available genetic data.
