OPTIMASI PERTUMBUHAN BAKTERI TERMOFILIK Bacillus tropicus DS2D UNTUK MENINGKATKAN DEKOLORISASI ZAT WARNA NAPHTHOL ASBO-BLUE B
Batik industry waste has the potential to cause significant environmental pollution, especially due to the use of synthetic dyes such as Naphthol ASBO-Blue B. This dye is toxic, difficult to decompose, contains heavy metals, and can pollute water, causing changes in pH, increased Biochemical Oxygen Demand (BOD) and Chemical Oxygen Demand (COD), and decreased dissolved oxygen concentration. Biodecolorization using microorganisms, especially thermophilic bacteria, is a promising alternative to overcome this problem. Thermophilic bacteria such as Bacillus tropicus DS2D have the potential to degrade synthetic dyes at high temperatures. This study aims to determine the optimum dye concentration, temperature, and pH for the growth of thermophilic bacteria Bacillus tropicus DS2D in decolorizing synthetic dyes Naphthol ASBO-Blue B and to determine the chromatogram pattern of the results of decolorization of Naphthol ASBO-Blue B dyes under optimum conditions using thin layer chromatography (TLC). This research was conducted from August to December 2023 at the Genetics and Biotechnology Laboratory, and the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The research stages consisted of making liquid cultures, selecting the ability of thermophilic bacteria to decolorize dyes, measuring the ability of dye biodecolorization, optimizing bacterial growth to increase the biodecolorization power of Naphthol ASBO-Blue B which consisted of optimizing dye concentration, optimizing temperature, and pH, then analyzing the products of dye biodecolorization. The results showed that Bacillus tropicus DS2D was able to decolorize Naphthol ASBO Blue B at a dye concentration of 200 ppm for 7 days, 50°C, and pH 6 with a cell count of 2.2 x 108 CFU/ml and the largest biodecolorization percentage of 86.13% and Bacillus tropicus DS2D showed the formation of new compounds marked by the formation of two new spots on the sample results of Thin Layer Chromatography (TLC).
