EFEKTIFFITAS BEBERAPA METODE STERILISASI TERHADAP TINGKAT KONTAMINASI EKSPLAN TULANG DAUN DUKU (LANSIUM DOMESTICUM) PADA KULTUR IN VITRO.
The Duku plant (Lansium domesticum) is a tropical fruit in indonesia that is in great demand by the public. Tissue culture is an alternative for producing high-quality duku plant seedlings in large quantities and in a short time, referring to the concept of the totipotency of plant cells, and capable of eliminating pests and diseases through the sterilization stage of the explants. This research was aimed to compared different materials and procedures on duku plant explants (Lansium domesticum) through tissue culture. This research was conducted at the Tissue Culture Laboratory, Department of Agronomy, Faculty of Agriculture, Sriwijaya University,with location coordinates (2°59’23.4”S104°43’53.4”E) The research was carried out from June to September 2024. The results showed that different combinations and duration of NaOCl sterilization methods affect the percentage of living duku leaf bone explants. The combination of sterilization methods in P3 consisting of liquid detergent for 3 minutes, bactericide 0.2% for 10 minutes, fungicide 0.2% for 10 minutes, NaOCl 1% for 15 minutes, and 70% alcohol showed the lowest percentage of browning with a percentage value of 23%, the lowest bacterial contamination of explants at 3%, and the highest percentage of live compared to other sterilization method treatments.
