The problem that was often faced in hatching java combtail was that during the gonad maturation of male and female, it sometimes did not occur at the same time. Efforts that can be made through the application of reproductive biotechnology are sperm preservation or storage (sperm cryopreservation). Cryopreservation was a storage technique using low temperatures up to -196oC. Extenders and cryoprotectants important role in the cryopreservation process. One of the extenders that can be used was honey. The research aimed to determine the best dose to use honey as an extender in the cryopreservation of java combtail sperm. This cryopreservation research was carried out at the Laboratory Balai Pembibitan dan Hijauan Pakan Ternak (BPHPT), Sembawa, and the preparation of cryomedia solution and rearing of java combtail broodstock at the Aquaculture and Experimental Pond Laboratory, Aquaculture Study Program, Fisheries Department, Faculty of Agriculture, Universitas Sriwijaya. This research was conducted on July-August 2024. This research used a completely randomized design (CRD) consisting of (P0) 100% ringer’s lactate solution, (P1) 0.25% honey + 99,75% ringer’s lactate solution, (P2) 0.5% honey + 99,50% ringer’s lactate solution and (P3) 0.75% honey + 99,25% ringer’s lactate solution. The research results showed that pre-cryopreservation java combtail sperm included a sperm volume of 0.5 mL, sperm pH 7, medium sperm consistency, sperm movement duration of 120 seconds with a motility percentage score of 5 (100%) and sperm viability of 89.50%. The results of post-cryopreservation research on java combtail sperm for 14 days of storage showed the best results in P3 (0.75% honey + 99,25% ringer’s lactate solution) with a sperm motility score of 4 (90%) and sperm viability of 79.50%. Water quality data during rearing of java combtail broodstock in this study was pH 6.0-7.5 and temperature 25.5-28.5oC.
