UJI AKTIVITAS ANTIOKSIDAN FRAKSI DAUN KACA PIRING (Gardenia jasminoides) DENGAN METODE DPPH, FRAP, DAN FTC
Kaca piring (Gardenia jasminoides) is a traditional plant that serves medicinal purposes to treat fever, cold, swelling, and external wounds, potential as a natural antioxidant. The aim of this research was to determine the antioxidant activity of each fraction and to compare the activeness. The research was conducted with fractionated maceration based on step gradient polarity (n-hexane, ethyl acetate, methanol). The fractions were analyzed for total phenolic and flavonoid contents by UV-Vis Spectrophotometer. Antioxidant activity of G. jasminoides leaves were determined by DPPH (2,2-diphenyil-1-picrylhydrazyl), FRAP (Ferric Reducing AntioxidantPower), and FTC (Ferric Thiocyanate) methods. The research showed the total phenolic contents of n-hexane, ethyl acetate, and methanol fraction respectively 8.17; 8.54; 11.57 mgGAE/g and total flavonoid contents 2.77; 6.31; 6.83 mgQE/g. Antioxidant activity determination by DPPH assay resulted to the IC50 value of n-hexane, ethyl acetate, and methanol fraction respectively 1834.90; 1026.38; 872.13 mg/L. Antioxidant activity determination by FRAP assay resulted to the value of formed Fe2+ in n-hexane, ethyl acetate, and methanol fraction respectively 75.10; 101.05; 131.52 mg/L. Antioxidant activity determination by FTC assay resulted to the fat peroxidation inhibition percentage in n-hexane, ethyl acetate, and methanol fraction respectively 32.72; 43.09; 52.53 %. Thus, the highest antioxidant activity resulted in methanol fraction compared to the other fractions and directly proportional to the total phenolic and flavonoid content of each fraction.
