EFEKTIVITAS MERKURI KLORIDA (HGCl2) PADA STERILISASI EKSPLAN DAUN KOPI (COFFEA Sp.) DALAM KULTUR IN VITRO
Coffee plants are plantation crops originating from the African continent, precisely from the country of Ethiopia in the 9th century. Plants tissue culture is a technique used to produce new plants quickly and efficiently. This technique involves the use of culture media such as agar or agar gel that contains nutrients required by the plant. This study was aimed to test the effectiveness of mercuric chloride (HgCl_2) in sterilizing explants of coffee (Coffea sp.) leaf blade in vitro culture and to determine the best the best combination of HgCl_2 oncentration and explant soaking duration in maintaining explant survival and reducing microbial contamination. This research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University, Bukit Lama, Ilir Barat I Subdistrict, Palembang City, South Sumatra, from December 2024 to January 2025. The sterilization treatment used five combinations of sterilants, namely liquid detergent, streptomycin sulfate, benomyl, 70% alcohol, and mercury chloride (HgCl_2) different in concentrations and soaking in combination durations. The research design used was descriptive quantitative observation by calculating the percentage of living explants, the percentage of browning explants, and the percentage of microbial contamination (bacteria and fungi). The results showed that treatment P5 (Liquid Detergent for 5' + Streptomycin Sulfate 2 g for 10' + Benomyl 2 g for 10' + 70% Alcohol for 3' + HgCl2 0.3% for 1') gave the best results with the percentage of living explants reaching 93% on the 31st day after inoculation, without any browning on the explants and microbial contamination of only 7% which came from fungal microbes without any bacteria.
