Kissing gourami (Helostoma temminckii) is a native freshwater widely distributed in Southeast Asia such as Thailand, the Philippines, and Malaysia. The varied distribution of kissing gourami in different environments or habitats can cause genetic diversity. The purpose of this study was to determine the morphological, morphometric and meristic characteristics, analyze the diversity of mitochondrial COI gene sequences, and determine the genetic distance and phylogenetic between kissing gourami species, as well as the water quality of kissing gourami habitat. This research was conducted in November 2024-February 2025. Fish and water samples were taken from Kelekar River and Lebung Ulak Kemang. DNA barcoding stages included DNA extraction, DNA amplification using PCR, electrophoresis, and COI gene sequencing. The COI mtDNA gene fragment was amplified at 50.8oC for 30 seconds with 35 cycles. The nucleotide length of kissing gourami from Kelekar River and Lebung Ulak Kemang was 544 base pairs (bp). Samples of fish from Kelekar River and Lebung Ulak Kemang had similar percentages of 99.06-100%. Samples (HSK1, HSK2 HSK3, HSK4, HSK5, HLK1, HLK3 and HLK4) had a genetic distance of 0.0000 (0%), however all the samples had a genetic distance value of 0.002 (0.2%) with HLK2. Phylogenetic tree construction resulted in all samples were grouped in the first subcluster. The water quality parameters of the two locations included temperature of 31.3-34.2oC, pH 3.3-6.4, DO 4.4-5.8 mg L-1, TDS 0.13-2.15 mg L-1, alkalinity 10-64 mg L-1 and ammonia 0.068-0.342 mg L-1.
