Crossbanded barb is a native species to Indonesia which is one of the natural riches of Indonesian waters spread across Kalimantan and Sumatra. This research aims to determine the COI gene sequence of mitochondrial DNA, the genetic distance and phylogenetic of crossbanded barb in the Musi River and Kelekar River, and the water quality of the crossbanded barb habitat. This research was carried out in April-October 2023. Fish samples (n=14) and water samples were taken from the Musi River and Kelekar River. DNA barcoding stages include DNA extraction, DNA amplification using PCR, electrophoresis, and COI gene sequencing. The COI mtDNA gene fragment was amplified at an annealing temperature of 50.9ºC for 30 seconds for 35 cycles. Crossbanded bard from the Musi River and Kelekar River had differences in morphology in the form of scales and morphometrics in the form of differences in the ratio between standard length and body height. The concentration of DNA samples from the Musi River and Kelekar River ranged from 58-405.8 ng µL⁻¹. Crossbanded barb from the Musi River had an identity percentage of 99.85% with P. waandersi and crossbanded barb from the Kelekar River had an identity percentage of 99.83% with P. bulu. The length of crossbanded barb nucleotide from Musi River and Kelekar River was 676 bp, the genetic distance was 4.4%, and the phylogenetics formed 2 subclusters with a bootstrap value of 79%. The water quality obtained in the Musi River and Kelekar River was temperature 29.2-31ºC, pH 6.1-6.8, dissolved oxygen 6.3-9 mg L⁻¹, brightness 18-255.5 cm, current speed 8 .16-21.14 cm s⁻¹, TDS 0.01-0.02 mg L⁻¹, ammonia 0.13-1.11 mg L⁻¹, and alkalinity 10-16 mg L⁻¹.
