Rhizophora apiculata Bl., is a type of mangrove plant that can be utilized, especially in the leaves which are rich in bioactive compounds. The benefits of R. apiculata leaves are their ability as antioxidant agents. Therefore, further efforts are needed to identify other compounds contained in R. apiculata leaves and their bioactivity. This study aims to determine metabolite profiling, compound bioactivity through GC-MS analysis and antioxidant levels using the DPPH method. GC-MS analysis data were searched using the PubChem, chEBI, KEEG, and Spectabase websites. The chromatogram of GC-MS results showed that there were 7 compounds detected, namely: 2-Propanol, 1,1'-oxybis-, 1-Propanol, 2-(2-hydroxypropoxy)-, 2-O-Methyl-D-mannopyranosa, n-Hexadecanoic acid, Octadecanoic acid, Supraene, and Phenol, 3,5-bis(1,1-dimethylethyl)- with a total abundance of compounds reaching 100%. The compound that has a dominant abundance of 88.25% is the compound 2-O-Methyl-D-mannopyranosa which is included in the monosaccharide class. The metabolite profile detected four compounds which are the main compounds, namely: 2-O-Methyl-D-mannopyranosa (88.25%) as antibacterial, Supraene (1.33%) and Octadecanoic acid (1.43%) as antioxidants, and 2-Propanol, 1,1'-oxybis- (0.93%) as antimicrobial and as a humectant which is a raw material for cosmetics. Tracing the leaves of mangrove R. apiculata has several classes of compounds such as ether, monosaccharide, fatty acid, triterpenoid, and phenol classes. Each compound has different bioactivity and has dominant bioactivity as an antioxidant, antimicrobial, anti-inflammatory, and anticancer so that it can be used as an ingredient in making herbal medicine. The antioxidant content in R. apiculata leaves with a standard antioxidant value of quercetin of 93.13 ± 17.29 ppm and a standard ascorbic acid of 56.78 ± 10.01 ppm.
