UJI AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL DAN FRAKSI DAUN GELAM (MELALEUCA LEUCADENDRA (L.) L.) PADA TIKUS JANTAN GALUR WISTAR YANG DIINDUKSI CCL4
Free radicals are dangerous because they can cause degenerative disease. Free radicals can be neutralized by antioxidants. Plants that has potential as an antioxidant is gelam (Melaleuca leucadendra (L.) L.). Gelam leaves contain phenolics, flavonoids, steroids, triterpenoids, tannins, and saponins that have pharmacological activity as antioxidants. The study aimed to determine the characteristics of ethanol extract, secondary metabolit content and the antioxidant activity of gelam leaf extract and fraction by measuring malondialdehyde levels, catalase enzyme activity, macroscopic picture and histopathology of CCl4-induced rat livers. The study used 6 groups, namely positive control (Vitamin C 10mg/kgBB), negative control (Na-CMC 1%), ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction at a dose of 100 mg/kgBB. The results showed that the characterization of the extract met the requirements of the Ministry of Health of the Republic of Indonesia. The mean result of MDA levels of the positive control , negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction respectively 0.734; 2,428; 1,227; 0,812; 1,138; and 1.792 nmol/mL. The average result of catalase enzyme activity respectively 29.425; 10,514; 23,360; 28,706; 24,920; and 18,735 U/ml.. The n-hexan fraction of gelam leaves had the best antioxidant activity because the effect was equivalent to that of a positive control group.
