UJI AKTIVITAS ANTIDIABETES EKSTRAK ETANOL DAUN KENCANA UNGU (RUELLIA TUBEROSA L.) TERHADAP TIKUS PUTIH JANTAN GALUR WISTAR YANG DIINDUKSI ALOKSAN
Kencana Ungu (Ruellia tuberosa L.) is one of the natural plant that contain secondary metabolite such as flavonoid, triterphenoid, polyphenol which has the antidiabetic activity. The purpose of this study is to determine the results of phytochemical screening, total flavonoid content, %PKGD value, effective dose (ED50) and pancreatic histopathological profiles of the ethanol extract of kencana ungu leaves. This research was divided into 6 groups, namely the normal group (0.5% Na-CMC suspension), positive control (1 IU/kgBW insulin), negative control (alloxan + 0.5% Na-CMC suspension) and treatment of the ethanol extract of kencana ungu leaves with variations in doses of 100, 150, and 200 mg/kgBW. Rats was induced with a dose 125 mg/kgBW alloxan until blood glucose ≥ 126 mg/dL, every groups was given treatments for 14 days. The results showed that the ethanol extract of kencana ungu contains flavonoid, tanin, steroid and saponin with total flavonoid content 34,040±0,128 mgQE/g. The percentage reduction in blood glucose levels at doses 100, 150, and 200 mg/kgBW respectively by 43,24%; 47,79%; 51.98%, it didn’t differed significantly from insulin (p>0.05). Pancreatic histopathological observations showed that 200 mg/kgBW dose had the best cell regeneration effect with damage rate of 1-25%. The ED50 of the ethanol extract of kencana ungu leaves is 176,65 mg/KgBW. The ethanol extract of kencana ungu leaves is effective in reducing blood glucose levels with the highest percentage at 200 mg/kgBW dose.
