The research aimed to study the effect of adding concentrations of gambier extract and gum arabic on the physical, chemical and functional characteristics of gambier toothpaste. This research used a Factorial Completely Randomized Design (FCRD) with two treatment factors and each treatment was repeated three times. Factor A is the addition of gambier extract concentration (1.5%, 3% and 4.5%) and factor B is the addition of gum arabic concentration (8.2%, 9.7% and 11.2%). The parameters observed in this study were functional characteristics (antibacterial, antioxidant activity and total phenol), physical (color and viscosity), and chemical (pH). The data obtained ware processed using Analysis of Variance (ANOVA), the treatments that had a real effect was further tested using the Honet Real Difference Test (HRDT). The results showed that treatment A (increasing the concentration of gambier extract) had a significant effect on the lightness, redness, yellowness, pH, antioxidant activity, total phenol and antibacterial values. Meanwhile, treatment B (addition of gum arabic concentration) had a significant effect on the yellowness, viscosity, pH and antioxidant values. The interaction of the two treatment factors had a significant effect on viscosity, antioxidant and antibacterial activity. The treatment A3B3 (gambir extract concentration 4,5% and gum arabic concentration 11,2%) is the best treatment based on 7,3 mm, antibacterial functional characteristics, antioxidant activity 126,42 µg/ml, and total phenol 194,84 mgGAE/g, L* color physical characteristics 60,40%, a* 8,94, b* 12,84, viscosity 45166,7 cps, and A3B1 treatment (gambir concentration 4,5% and concentration of gum arabic 8,2%) are the best treatments based on pH 8,97chemical characteristics.
