Analysis of the genetic diversity of thermophilic bacteria is important to understand the species variation and functional potential possessed by bacteria. One One commonly used approach to identify and classify bacteria is DNA-based genetic analysis, specifically using the 16S rRNA gene, because it is sustainable and universal with identical functions in all bacteria. Research on “Genetic Diversity Analysis of Thermophilic Bacteria from Air Putih Hot Springs Lebong Bengkulu Using 16S rRNA Gene” aims to analyze the level of genetic diversity of thermophilic bacteria and determine the kinship relationship between identified thermophilic bacteria. Phylogenetic relationship analysis was carried out using MEGA 11 application based on neighbor-joining algorithm with bootstrap value of 1000 repetitions. Genetic diversity analysis using the DnaSp 5.10. The isolation results obtained 8 isolates of thermophilic bacteria, namely AAL1, AAL2, AAL3, ABL2, ABL3, SCL1, SCL2, and SCL3, with characteristics that are bacillus-shaped, have gram-positive properties and produce endospores. The level of genetic diversity of eight isolates of thermophilic bacteria is included in the category of high genetic diversity with each value in each isolate in order; isolates AAL3, ABL2, ABL3, SCL2, and SCL3 value of 0.977; isolates AAL1 and AAL2 each value is 0.933; and isolate SCL1 value is 0.800. Based on the phylogenetic tree, it can be assumed that isolate AAL1 is closely related to Bacillus sonorensis strain NBRC 101234; isolate AAL2 has a close relationship with Bacillus mojavensis strain NBRC 15718; isolates AAL3, ABL2, ABL3, SCL2 and SCL3 have a close relationship with Bacillus licheniformis strain ATCC 14580; and isolate SCL1 has a close relationship with Bacillus paralicheniformis strain KJ-16.
