VARIASI GENETIK TARSIUS ASAL PULAU BELITUNG BERDASARKAN GEN PENANDA DISPLACEMENT LOOP (D-LOOP)
Land clearing on Belitung Island has caused significant environmental impacts. The existence of the Belitung tarsier is currently quite alarming, because its status has been declared as Endangered or threatened and has not received adequate attention in terms of genetic conservation, so its genetic information is still very limited. To address this issue, further research is needed on analyzing genetic variation in Belitung tarsier individuals using Displacement Loop (D-Loop) genetic markers. This study aims to analyze genetic variation among Belitung tarsier individuals using the Displacement Loop (D-Loop) gene marker. The research samples used were ear tissue samples. The samples used came from the Bukit Peramun location, Air Selumar Village, Belitung and from the Batu Mentas location in Kelekak Datuk Village, Belitung. The research stages consisted of total DNA isolation, quantity test and quality test, DNA amplification, electrophoresis, sequencing, data analysis using GeneStudio application, BLAST NCBI website, Mega program and DNAsp. DLTARPROF and DLTARBFR primers successfully amplified all six samples indicated by the presence of DNA bands ranging from 418 bp to 424 bp. The Hd value is 1 in each population so it is concluded that the six samples have a very high level of genetic variation based on the Displacement Loop (D-Loop) marker gene.
