The abundance of keratin in nature leads to serious problems because keratin is a type of protein that is difficult to degrade. Keratinase, a specific type of proteolytic enzyme, is used as an alternative for keratin waste management, with thermophilic bacteria as potential organisms for producing thermally stable keratinase enzymes. One of the hot springs located in North Lebong District, Lebong regency, Bengkulu features a hot spring with temperatures of 100-115℃, which allows for the growth of thermophilic bacteria. However, no research has been conducted at this hot spring, resulting in a lack of information about thermophilic bacteria in the area. The isolation and molecular identification of thermophilic bacteria from the hot water source in Air Putih Lebong, Bengkulu is necessary to obtain thermophilic bacteria with the potential to produce keratinase. The objective of this research is to acquire thermophilic bacteria capable of producing keratinase and to understand the phylogenetic relationships of these thermophilic bacterial species. This study will be conducted from August 2024 to December 2024 at the Genetics and Bitechnology Laboratory, Microbiology Laboratory, Department og Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The research stages include the selection of keratinolytic bacteria, macroscopic and microscopic characterization of the bacteria, molecular identification of the bacteria, and data analysis and presentation. Based on the result, four isolates were found to have keratinase activity, indicated by the formation of clear zones around the bacterial colonies growing on keratin media. The morphological characteristics of the colonies obtained were regular in shape, with colony colors ranging from white to yellowish-white, exhibiting umbonate and flat elevations with varied edges. The morphological characteristics of the cells revealed that the bacteria are gram positive with a bacillus shape and possess endospores. The fourth isolate, after identification, was found to have the closest phylogenetic relationship with Bacillus licheniformis strain ATCC 14580 and Bacillus mojavensis strain NBRC 16718.
