OPTIMASI FORMULASI NANOPARTIKEL EKSTRAK DAUN KENIKIR (Cosmos caudatus Kunth.) BERBASIS POLIMER KITOSAN-ALGINAT TERKONJUGASI ASAM FOLAT
Kenikir leaves (Cosmos caudatus Kunth.) contain flavonoid group compound quercetin which has potential as anti-cancer. The extract of kenikir leaves has low solubility and bioavailability so that a modification of the drug delivery system is needed in the form of folic acid-conjugated chitosan-alginate polymer-based nanoparticles. Folic acid-conjugated chitosan carries the drug to folate receptors that are overexpressed in cancer cells. The purpose of this study was to obtain the best optimum formula and determine the results of nanoparticle characterization. The polymer concentration used with variation of chitosan and alginate concentration F1 (0.3% and 0.3%), F2 (0.3% and 0.6%), F3 (0.6% and 0.3%), and F4 (0.6% and 0.6%). The extract of kenikir leaves produced a yield of 25.03% and the results of phytochemical screening contained flavonoids, tannins, triterpenoids, steroids, alkaloids, and saponins. Extract characterization obtained a moisture content of 7.40%, water soluble juice content of 45.60%, ethanol soluble juice content of 74.16%, and total ash content of 3.60%. The results of conjugate characterization with FTIR showed successful conjugation characterized by the appearance of stable amide bonds from folic acid and chitosan. The amount of folic acid that was successfully conjugated was very high at 99.49%. Nanoparticle characterization of all formulas had sizes in the range of 145-450 nm, PDI 0.305-0.481, zeta potential -18 to -19 mV, and %EE 88-92%. The optimum formula obtained chitosan concentration of 0.3% and alginate concentration of 0.6% with a particle size of 132.2 nm, PDI of 0.266, zeta potential of -29.4 mV. TEM morphology test of nanoparticles has irregular round shape and uniform distribution. The characterization results of the optimum formula of the cycling test obtained decreased %EE from 93.761% to 88.956% and decreased pH from 7.4 to 7.3 but the nanoparticle preparation is still said to be stable. Based on the test results obtained, the optimum formula produces a stable nanoparticle preparation characterization at an alkaline pH of 6-7.4, chitosan is successfully conjugated with folic acid for cancer cell targeting, and the drug was released well with a drug release percentage of 83.01%.
