PRODUKSI ENZIM SELULASE DARI ASPERGILLUS NIGER MENGGUNAKAN SUBSTRAT AMPAS TEBU
A Study Has Been Conducted To The Production Of Cellulase Enzyme From Aspergillus Niger Using Bagasse Substrate. Bagasse Is A Lignocellulosic Waste That Contains High Lignin So It Needs To Be Delignified To Remove The Lignin Content. Lignin Content Can Be Determined By The Chasson-Datta Method. The Delignification Process Succeeded In Reducing The Lignin Content By 71.84%, From 22.77% To 6.41%, Making Bagasse Better Used For The Fermentation Process By A. Niger. The Delignified Bagasse Was Used As A Substrate For Fermentation Of Cellulase Enzyme Production Using A. Niger. Cellulase Enzyme Activity From The Fermented Crude Extract Was Tested By Dns Method And Protein Content Determination By Lowry Method. Based On The Results Of The Study, It Was Found That The Production Of Cellulase Enzyme On Bagasse Substrate With The Help Of A. Niger Fungus Was Able To Work Optimally At Ph 5, With A Temperature Of 35°C And A Substrate Concentration Of 2% Cmc Which Resulted In A Cellulase Enzyme Activity Of 0.01434 U/Ml. Based On The Lineweaner-Burk Equation, The Vmaks Value Of 0.022 U/Ml And Km Value Of 1.724 Mg/Ml Were Obtained.
