Red onion skin (Allium cepa L.) has been proven to promote hair growth due to its strong antioxidant activity. Transfersomes were capable of penetrating the transfollicular pathways to enhance the antioxidant effect of quercetin compounds in red onion skin extract. This study aims to optimize the transfersome formula of red onion skin extract by varying the concentrations of phospholipon 90G and tween 80 using a 2² factorial design method in Design-Expert 13®, evaluating the responses of encapsulation efficiency, polydispersity index, particle size, and zeta potential. The optimal transfersome formula was developed in a dispersion system and underwent evaluation of its preparation, morphology test and antioxidant activity assays. Based on the analysis results the optimal transfersome formula was obtained with a phospholipon 90G to tween 80 concentration ratio of 85:15. The optimum formula showed brick-red color and no sediment. The encapsulation efficiency, particle size, polydispersity index, and zeta potential exhibited favorable results, with respective values of 96,3971±0,0204%, 100.996±0.750 nm, 0.212±0.012, and -8.616±0.508 mV. The characterization results of the transfersome dispersion system revealed a viscosity of 601.7625 cps. The dispersion system exhibited a brick-red color, homogeneous, and showed adhesion times of 4.16 seconds (50 g) and 4.50 seconds (100 g). The spreading distances were measured at 6.53 cm (50 g), 7 cm (100 g), and 7.4 cm (150 g). The pH was measured at 6.57, and the quercetin content was determined to be 114.0404 mg QE/g of extract. The transfersomes were spherical shape, and the transfersome dispersion system exhibits strong antioxidant activity (IC50 65.224 ppm).
