Gamal leaves (Gliricidia sepium) have pharmacological activity as anthelmitic, hepatoprotector and antibacterial. The content of secondary metabolites in the ethyl acetate fraction of gamal leaves are flavonoids, tannins and saponins. The purpose of this study was to determine the effects of acute toxicity of the ethyl acetate fraction of gamal leaves with testing parameters of biochemical levels, macroscopic and microscopic images of liver, kidney and heart organs on male white rats wistar strain. In this study using the fixed dose procedure method. In the preliminary test, the test animals were divided into 5 groups, namely normal (Na-CMC 0.5%) and dose variations of 5, 50, 300, and 2000 mg/kgBB given orally for one day. The main test was divided into 2 groups, namely the normal group and the 2000 mg/kgBB dose group with each group consisting of 5 test animals for 14 days. ased on the preliminary test results, no test animals died. This study was continued in the main test and obtained the average value of biochemical parameter levels in the normal group, namely SGOT 116.52 ± 14.54 U/L, SGPT 81.40 ± 5.89 U/L, creatinine 0.63 ± 0.08 mg/dL, ureum 36, 63±5.25 mg/dL, while in the 2000 mg/kgBB dose group obtained SGOT 114.25±6.78 U/L, SGPT 84.80±3.19 U/L, creatinine 0.70±0.03 mg/dL, and ureum 33.52±5.83 mg/dL had no significant effect (p>0.05). The ethyl acetate fraction of gamal leaves (Gliricidia sepium) at a dose of 2000 mg/kgBB had no effect on the macroscopic organs of the liver, kidneys, and heart. The microscopic picture of the liver and kidney organs experienced a moderate degree of damage (multifocal) and the heart organ experienced a mild degree of damage (focal). Administration of the ethyl acetate fraction of gamal leaves did not cause toxic symptoms or death in the 2000 mg/kgBB dose group.
